have you tried disabling tooltips? that's the only click-related bug that i know of
EDIT: also i'm assuming you're not talking about the actual Click Buffer option in character prefs? I play with that toggled On, tooltips Off and never have any problems with clicks being ignored

yeah theres a bug with tooltips where sometimes you have to click at least twice to actually do things. just turn them off or set them to ALT only if you haven't already
